Ipamorelin is a synthetic pentapeptide growth hormone secretagogue, Aib-His-D-2-Nal-D-Phe-Lys-NH2. Scientists at Novo Nordisk reported it in 1998 (Raun et al., PMID 9849822). GSRS and ChEMBL list NNC 26-0161 as its code name. It was found in a series derived from GHRP-1 that lacks the central Ala-Trp dipeptide. It releases growth hormone through a GHRP-type receptor, the growth hormone secretagogue receptor type 1a (ghrelin receptor). In swine it did not raise ACTH or cortisol, unlike GHRP-6 and GHRP-2. Development reached phase 2 for post-operative ileus. In the published phase 2 trial (117 patients, intravenous dosing, PMID 25331030) the median time to the first tolerated meal was 25.3 h with ipamorelin and 32.6 h with placebo, and the difference was not statistically significant. Development then stopped, and no regulator has approved it.

Differences from the parent
Basis: Pharmacological analogy only. Ipamorelin agonises the same receptor as ghrelin (growth hormone secretagogue receptor type 1a; GSRS relationship TARGET->AGONIST). Its sequence is not derived from ghrelin: the ipamorelin residues share no segment with ghrelin-28 (UniProt Q9UBU3, residues 24-51), which contains no tryptophan or naphthylalanine. Treat this link as a family link, not a sequence relation.
Calculated from the two sequences with @pepdb/chem alignToParent (ungapped unless a gap scores higher). The record’s curated notes are shown as given.
Some values cannot be calculated. The sequence has a non-canonical residue (X). Residue tables cover only the 20 standard amino acids.
