Cibinetide, research code ARA-290, is an 11-residue peptide that copies the water-facing side of helix B of erythropoietin. Brines and colleagues reported in 2008 that this surface carries the tissue-protective activity of erythropoietin, which is mediated by a receptor heterocomplex of the erythropoietin receptor and CD131 (the innate repair receptor), while red-cell production is mediated by the erythropoietin receptor homodimer; the peptide is not erythropoietic in vitro or in vivo. Araim Pharmaceuticals developed it for small nerve fibre loss in sarcoidosis and for islet transplantation, with orphan designations in the United States and the European Union. Development has reached phase 2.

Differences from the parent
Basis: Designed from the three-dimensional structure of erythropoietin, not cut from its sequence. Brines et al. 2008 report that helix B (residues 58-82) faces the aqueous medium when erythropoietin is bound to the receptor homodimer and that an 11-residue peptide built from the amino acids forming that aqueous face is tissue protective and not erythropoietic. A substring check against the mature EPO chain confirms the cibinetide sequence is not contiguous in EPO.
Calculated from the two sequences with @pepdb/chem alignToParent (ungapped unless a gap scores higher). The record’s curated notes are shown as given.
Some values cannot be calculated. The sequence has a non-canonical residue (X). Residue tables cover only the 20 standard amino acids.
