Mazdutide (IBI362, LY3305677) is a synthetic 34-residue oxyntomodulin analog with Aib at position 2, a C20 fatty diacid side chain on Lys20 and a C-terminal amide. It is an agonist at the GLP-1 receptor and the glucagon receptor. Innovent Biologics develops it in China under license from Eli Lilly; Innovent reports NMPA approval for chronic weight management (2025-06-27) and for glycemic control in type 2 diabetes (2025-09-19). Lilly has run phase 2 studies in the United States; it is not approved by FDA or EMA.

Differences from the parent
Basis: KEGG D12225 classes mazdutide as an oxyntomodulin analog (DG03355). Substitutions are computed for this record by an ungapped position-by-position comparison of KEGG D12225 with UniProt P01275 residues 53-89. Mazdutide has 34 residues; oxyntomodulin residues 35-37 (NIA) have no counterpart.
Calculated from the two sequences with @pepdb/chem alignToParent (ungapped unless a gap scores higher). The record’s curated notes are shown as given.
Some values cannot be calculated. The sequence has a non-canonical residue (X). Residue tables cover only the 20 standard amino acids.
@pepdb/chem 0.2.0 · EMBOSS pKa set · ExPASy residue masses
No charge curve
The sequence has a non-canonical residue (X). Residue tables cover only the 20 standard amino acids.
34 residues. Each residue counts in one class; F and W count as aromatic.
Loads the 3D viewer (3Dmol.js, about 150 KB) and the model file from AlphaFold DB (EMBL-EBI).
An ideal alpha-helix projection. It shows where the hydrophobic residues fall if the peptide folds as a helix. It does not show that it does.
Eisenberg consensus (normalized) scale · + K, R · − D, E
Assumes an ideal α-helix: 100° per residue, 3.6 residues per turn. The arrow is the mean hydrophobic moment (Eisenberg et al. 1984) and points at the hydrophobic face.
