Survodutide (BI 456906) is a 29-residue synthetic peptide designed by adding GLP-1 receptor activity to the glucagon sequence (Zimmermann et al. 2022). It has 1-aminocyclobutane-1-carboxylic acid (Ac4c) at position 2, a C-terminal amide and a C18 fatty diacid on Lys24, attached through L-gamma-Glu and a Gly-Ser-Gly-Ser-Gly-Gly linker (GSRS). It is a glucagon receptor and GLP-1 receptor dual agonist. Boehringer Ingelheim develops it under license from Zealand Pharma; it is in phase 3 trials for obesity and MASH, and no approval was found in Drugs@FDA or the EMA data on 2026-10-09.

Differences from the parent
Basis: Zimmermann et al. 2022 (Mol Metab, PMID 36356832): BI 456906 was designed 'by introducing GLP-1R activity into the glucagon amino acid sequence'. Substitutions are a direct alignment with human glucagon (UniProt P01275, residues 53-81). Survodutide also has a C-terminal amide; glucagon has a free acid.
Calculated from the two sequences with @pepdb/chem alignToParent (ungapped unless a gap scores higher). The record’s curated notes are shown as given.
Some values cannot be calculated. The sequence has a non-canonical residue (X). Residue tables cover only the 20 standard amino acids.

No charge curve
The sequence has a non-canonical residue (X). Residue tables cover only the 20 standard amino acids.
29 residues. Each residue counts in one class; F and W count as aromatic.
Loads the 3D viewer (3Dmol.js, about 150 KB) and the model file from AlphaFold DB (EMBL-EBI).
Survodutide has 29 residues. A full-atom drawing is hard to read at this size; the sequence view is the reference.
